The Saint Antoine Research Centre (CRSA), INSERM / Sorbonne University, is offering a two-year postdoctoral fellowship in adipose tissue pathophysiology.
The successful candidate will work on the metabolic sub-study of the international ELDORADO clinical trial, which compares the effects of two HIV antiretroviral treatment regimens on adipose tissue distribution and function.
The postdoctoral researcher will join the team “Lipodystrophies, metabolic and hormonal adaptations, and aging”, led by Pr B. Fève, and will work with Pr V. Béréziat and Dr C. Lagathu, with Pr J. Capeau as emeritus professor.
🔬 Research Project
The project focuses on understanding the effects of antiretroviral treatment on adipose tissue phenotype, distribution, morphology, and function in people with HIV.
The research will involve:
- Characterization of adipose tissue phenotype from biopsies.
- Analysis of gene expression and global transcriptome.
- Investigation of adipose tissue function.
- Comparison of doravirine/tenofovir/lamivudine and dolutegravir/tenofovir/emtricitabine or lamivudine treatment regimens.
- Analysis of adipose tissue quantity and quality.
- Evaluation of adipose tissue distribution, morphology, and histology.
- Investigation of mechanisms underlying antiretroviral-associated weight gain.
- RNA-seq and transcriptomic analysis.
- Preclinical validation of mechanisms identified through the clinical study.
🧪 What You’ll Work On
1️⃣ Adipose Tissue Characterization
The postdoctoral researcher will characterize adipose tissue samples obtained from participants with HIV initiating antiretroviral treatment.
Key activities will include:
- Analysis of adipose tissue phenotype.
- Gene expression studies.
- Evaluation of tissue function.
- Histological and morphological characterization.
- Investigation of adipose tissue quantity and quality.
2️⃣ Molecular & Cellular Biology
The project will involve molecular and cellular approaches to investigate treatment-associated changes in adipose tissue.
Techniques may include:
- qPCR.
- Western blotting.
- Immunofluorescence.
- Cellular and molecular biology techniques.
- Imaging and tissue analysis.
3️⃣ Transcriptomics & RNA-seq
The researcher will contribute to the analysis of global transcriptomic changes associated with antiretroviral treatment.
This will include:
- RNA-seq analysis.
- Gene-expression profiling.
- Identification of molecular pathways associated with weight gain.
- Interpretation of transcriptomic data.
4️⃣ Comparative Antiretroviral Analysis
The project will compare the effects of different antiretroviral regimens on adipose tissue, particularly focusing on:
- Adipose tissue distribution.
- Tissue morphology.
- Histology.
- Tissue dysfunction.
- Metabolic consequences.
- Treatment-associated weight gain.
5️⃣ Preclinical Validation
Mechanisms identified through the ELDORADO clinical trial will be further investigated through preclinical approaches to understand the biological mechanisms underlying antiretroviral-associated weight gain.
🎯 Ideal Candidates
Applicants should have a PhD in cell biology, molecular biology, biochemistry, or a related field, with strong practical skills in cellular and molecular biology.
Candidates should possess:
- A PhD in cell biology, molecular biology, biochemistry, or a related discipline.
- Strong experience in cellular and molecular biology techniques.
- Practical laboratory research experience.
- Experience with molecular and cellular analysis.
- Ability to work independently and as part of a multidisciplinary research team.
- Strong interest in adipose tissue biology, metabolism, or pathophysiology.
